Dexlansoprazole (中文: 右蘭索拉唑)
Drug Class[edit]
Proton Pump Inhibitors (PPI)
Common Strengths of Dexlansoprazole capsule[edit]
Dexlansoprazole capsule are available in the following strengths:
- 30 mg delayed-release capsules
- 60mg delayed-release capsules
Drug Trade Names of Dexlansoprazole capsule[edit]
Dexilant
Drug Usage[edit]
Dexlansoprazole is used for:
- Healing of erosive esophagitis (EE)
- Maintenance of healed erosive esophagitis (EE)
- Treatment of gastroesophageal reflux disease (GERD)
Mechanism of Action[edit]
Inhibits (H+/K+ )-ATPase enzyme in the gastric parietal cells, blocking gastric acid production in the stomach
Route of Administration[edit]
Oral
Dosages[edit]
For adults and children aged 12 years and older, the typical dosage is:
Healing: 60 mg once daily for up to 8 weeks
Maintenance: 30 mg once daily for up to 6 months
GERD: 30 mg once daily for up to 4 weeks
Renal Dose Adjustments[edit]
No renal dose adjustment recommended
Side Effects[edit]
| Frequency | Adverse reactions |
|---|---|
| Common Side Effects | |
| Diarrhea | |
| Abdominal pain | |
| Nausea | |
| Vomiting | |
| Flatulence | |
| Serious Side Effects | |
| Bone fractures | |
| Low blood magnesium levels | |
| Clostridium difficile infection | |
| Anaphylaxis | |
| Vitamin B12 deficiency |
Pharmacokinetics[edit]
Dual-release mechanism
Onset of action: 1-2 hours (first peak), 4-5 hours (second peak);
Duration of action: Up to 24 hours, longer than other PPIs
Drug Precautions[edit]
Pregnancy: Caution advised; Children: Safety not established; Elderly: Monitoring recommended
Breastfeeding: safety during breastfeeding is unclear
Children and Elderly: Approved for use in children aged 12 years and older; elderly patients may require monitoring due to potential increased sensitivity.
Monitoring Items[edit]
Regular monitoring of magnesium and vitamin B12 levels
Drug Interactions[edit]
Dexlansoprazole can interact with drugs metabolized by CYP2C19 and CYP3A4 enzymes
Interacts with methotrexate, digoxin, warfarin, certain antiretrovirals
